I am swapping my autolite 4300 with a Holley 4150 600cfm with electric choke.
i removed my tube from the choke to the exhaust manifold, how should i plug the hole?

I don't think you'll hear any significant exhaust leak nor have any measurable soot come through it, but a plug with some anti-sieze may help preserve the opening from corrosion if you or someone else decided to return it to stock.

The choke tubes attach to a heat stove, and do not connect to exhaust in any way.

thanks for the replies, I wasnt sure if exhaust was going through it since it was very clean.
I replaced back in 1998. it pulled out and was only a little dirty looking.

The hole is not connected to the exhaust gas portion of the manifold. It is a way to pull outside air through a hole in the manifold to warm it, not to pull exhaust gas into the air intake.
